RESUMO

OBJECTIVE@#To investigate the expression of Talin1 in the fallopian tube and chorionic villi in patients with tubal pregnancy and its role in regulating invasion and migration of trophoblasts.@*METHODS@#Immunohistochemistry and Western blotting were used to detect the localization and expression level of Talin1 in the fallopian tube and chorionic villi in patients with tubal pregnancy and in women with normal pregnancy. In the cell experiment, HTR-8/SVneo cells was transfected with Talin1 siRNA and the changes in cell invasion and migration were assessed using scratch assay and Transwell assay. The expressions of MMP-2, MMP-9, N-cadherin and Snail in the transfected cells were detected by qRT-PCR and Western blotting.@*RESULTS@#Positive expression of Talin1 was detected in both normal fallopian tube tissues and tissues from women tubal pregnancy, and its expression was localized mainly in the cytoplasm of cilia cells. The expression level of Talin1 was significantly higher in both the fallopian tube and chorionic villi in women with tubal pregnancy than in normal fallopian tube and chorionic villi samples (P < 0.01). In HTR-8/SVneo cells, transfection with Talin1 siRNA significantly inhibited cell invasion (P < 0.01) and migration (P < 0.05), down-regulated the expression of N-cadherin, MMP-2 and Snail (P < 0.05), and up-regulated the expression of MMP-9 in the cells (P < 0.05).@*CONCLUSION@#The expression of Talin1 in the fallopian tube and chorionic villi is significantly increased in women with tubal pregnancy, suggesting the association of Talin1-regulated trophoblast cell invasion with the occurrence of tubal pregnancy.

RESUMO

Depression is a psychiatric disease that seriously affects human life in the context of rapid social development and increased living pressure, and brings serious healthproblems and economic burdens to human society.Traditional Chinese medicine has a long history in the treatment of depression, and Xiaoyaosan has been repeatedly confirmed as a classic prescription for the intervention in depression, but its antidepressant mechanism and its active ingredients are still unclear.Through the search of China Knowledge Network(CNKI) and PubMed database, the literature and data on the clinical and mechanism research of Xiaoyaosan and its analogous prescriptions on depression in the past 10 years are systematically sorted out.It is found that Xiaoyao San and its analogous prescriptions are mainly focusing on the following aspects: regulating hypothalamic-pituitary-gonad, hypothalamic-pituitary-thyroid, and hypothalamic-pituitary-adrenal axis, affecting 5-hydroxytryptamine(5-HT), norepinephrine(NE) neurotransmitters in hippocampus, interfering with inflammatory reaction process, modulating the BDNF/CREB signaling pathway, regulating intestinal flora and gastrointestinal function, regulating endothelial window foramen of hepatic sinus, repairing chronic stress-induced blood-brain barrier permeability, affecting autophagy to reduce neuronal apoptosis, and regulating the leukoplakia-nuclear adrenal gland (LC-NE) system to play an antidepressant role.The main active ingredient including, saikosaponin A which could down-regulate interleukin expression level, saikosaponin D which alleviates HPA axis dysfunction, quercetin which inhibits corticotropin-releasing-factor(CRF) mRNA expression, paeoniflorin which regulates neurotransmitters, and paeoni lactone which inhibits hyperfunction of HPA axis Glycosides, reduce inflammatory cytokines, atractylenolide I, atractylenolide Il, atractylenolide Ill, which could reduce the expression of pro-inflammatory cytokine mRNA and inhibit NF-κB activated, curcumin which could play the neuroprotective and anti-inflammatory and antioxidant roles, L-menthol which could suppress the HPA axis overactivation, liquiritin isoliquiritin which could regulate the monoamine neurotransmitters, and isoliquiritigenin which could inhibit the monoamine oxidase.

RESUMO

With the standardized training of resident doctors, the importance of clinical practice becomes more and more important. As the key department of clinical practice, the gynecology department of traditional Chinese medicine pays more attention to the training of clinical thinking. The implementation of scene reproduction mode combined with case discussion mode in clinical practice teaching of TCM postgraduates has improved the quality of clinical training, strengthened the transformation of clinical knowledge, expanded the train of thought of scientific research, and achieves good education results.

RESUMO

Pharmacological activities of the effective constituents: essential oils, flavonoids andalkaloids, in fruit of Citrus aurantium L. were compared. Blood pressure was increased markedly by transfusion of the alkaloids. Contractions of isolated intestines were iuhibited byflavonoids. Essential oils significantly decreased the rate and frequency of writhing syndromeinduced by ip acetie acid and spontaneous locomotor activities in the mice with the exceptionfexciting at first followed by inhibiting the isolated intestines,which displayed analgesia andoceutral inhibition to a certain extent.
